This place is included on the Victorian Heritage Inventory, for its potential to contain historical archaeological remains associated with the settlement and growth of early Melbourne. Under the terms of the Heritage Act 2017 there is protection for all historical archaeology sites and objects in the state.

Hotel constructed on site 1850, shown 1855 map - Duke of Wellington.1877 - 2 & 1 storey buildings, outbuildings, yard; Duke of Wellington Hotel.1888 - 2 storey building, yard, outbuilding, Duke of Wellington Hotel; 3 shops, A Stern, Draper.1905 - same buildings, A Stern, Draper; A Pullinger, Stove Maker.
Heritage Inventory Significance: CITATION - NOTABLE BUILDING (AM425): THE CORNER SECTION OF THE DUKE OF WELLINGTON HOTEL WAS ORIGINALLY BUILT AS A STONE HOUSE FOR A TIMOTHY LANE IN 1850. IT WAS LICENSED AS A HOTEL IN 1853. IT WAS LATER EXPANDED TO INCORPORATE THE ADJOINING BUILDINGS AND GIVEN A UNIFORM EXTERNAL TREATMENT. IT IS THE OLDEST HOTEL STILL OPERATING IN MELBOURNE.
